The word histology originally meant "the study of
science of tissues" although now, it is taken to mean "the study
of the fine structure of animal tissues and organs". The basis of
good histological preparations is adequate and complete fixation or preservation
of the tissues. Fixation is necessary to prevent post mortem changes ie.
autolysis, putrefaction and to preserve the tissue in as life like a manner
as possible. It also hardens the naturally soft tissues so that they will
be easily handled during the subsequent treatments and leaves the tissues
in a state which facilitates differential staining with dyes and other
reagents.
